Gleason Nyu-York shahrida Mina va William L. Gleasonning oʻgʻli sifatida tugʻilgan[3]. Teatrdan kelib, maktab oʻquvchisi sifatida taʼtil paytida sahnaga chiqdi. U oʻn uch yoshida xabarchi, matbaachi, elektr doʻkonida yordamchi va liftchi boʻlib tirikchilik qila boshlagan. 16 yoshida Qoʻshma Shtatlar armiyasiga qoʻshilgan va Filippinda uch yil xizmat qilgan.
Ishdan boʻshatilgandan soʻng u oʻzining sahna karerasini boshladi, keyinchalik uni professional tarzda davom ettirdi. U Londonda ikki yil rol oʻynadi va Qoʻshma Shtatlarga qaytganidan soʻng komediyalar uchun dialoglar yozish orqali filmlarda qatnashdi. U bir qancha pyesalar yozgan, ulardan bir nechtasi Brodveyda ijro etilgan. Shuningdek, Brodveyda oʻzining bir nechta pyesalarida rol oʻynagan.
